A Research on “Formulation & Evaluation of Mouth Dissolving Tablet of Azithryomycine Dihydrate”

Azithromycin dihydrate is macrolide antibiotic. It is widely used in the treatement of upper and lower respiratory trac infection, prevention of disseminated Mycobacterium avium complex (MAC) infection in patients with advanced human immunodeficiency virus (HIV) infections &uncomplicated skin and skin structure infections like Folliculitis, Cellutis, and Erysipelas. Although it is commonly used but the major problem is its bitterness. Oral administration constitutes the preferred route for administering Azithromycin dihydrate. Due to the decline in the swallowing ability with age, elderly patients complain that it is difficult for them to take some currently used dosage form such as tablets and capsules. For this reason the tablets that can rapidly disintegrated in the mouth or in the oral cavity were developed. Instead of disintegrating in water (Dispersible tablet), mouth dissolving/disintegrating tablet disintegrate in the oral cavity without drinking water. The disintegrated mass can slide down smoothly in the esophagus with the help of saliva. So even people who have swallowing or chewing difficulties can take it with ease. Moreover these tablets have sufficient mechanical integrity to able to withstand handling without breakage.In the present work, taste kasking of Azithromycin dihydrate was carried out by using Eudragit E 100 (mass extrusion method). These taste masked granules complex was further formulated into the mouth disintegrating tablet by direct compression method using Crospovidone, Sodium starch glycolate &Ac-di-sol as a superdisintegrant. When it comes in contact with saliva or gastrointestinal fluid, causing rapid disintegration without the formation of lumps. It also provides necessary hardness and chemical stability to the tablet.
